After lyophilization, the dried product is often sealed under vacuum or an inert gas to limit moisture and oxygen exposure. Vials, stoppers, and seals must maintain their barrier throughout shelf life. Storage temperature depends on product sensitivity: some cakes tolerate controlled room temperature, while labile biologics require refrigeration. Humidity is a critical variable because dried cakes are hygroscopic and can absorb water when exposed to air. Handling procedures therefore limit open-vial time and use desiccated environments for sampling.
Quality control for lyophilized materials examines appearance, reconstitution time, residual moisture, and mechanical integrity. An acceptable cake is usually uniform and porous, though appearance alone does not prove stability. Karl Fischer titration is a common method for water content, while differential scanning calorimetry can reveal glass transition events. Stability studies track potency, aggregation, and moisture over time under defined temperature and humidity conditions. Specifications are product-specific and may include sterility and endotoxin tests for sterile preparations.
Misconceptions about lyophilization include the idea that dried products are indefinitely stable or that vacuum sealing eliminates all degradation. Chemical reactions can continue in the solid state, and some proteins lose activity even at low moisture. Another misconception is that any freeze-dryer cycle can be scaled by time alone; heat and mass transfer differ with equipment and load. Open questions remain about predicting long-term stability from short accelerated studies, particularly for amorphous formulations. Real-time stability data remain the standard for shelf-life assignment.
Lyophilization removes water by freezing a material and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intermediate liquid state. Because the material remains frozen during primary drying, the structure often stays porous. This porous matrix can rehydrate quickly when water is added back. The low pressure also allows vapor to leave the solid matrix without boiling.
A typical cycle begins with freezing, which fixes the material into a solid and determines ice crystal size. Primary drying then raises heat under vacuum so ice sublimes, often near or below the collapse temperature of the formulation. Secondary drying removes bound water that remains after ice is gone, usually by gently warming the product. Each stage balances heat input against pressure to avoid melting or structural damage. Temperature probes and pressure sensors guide the transition between stages.
In practice, lyophilization is slower and more energy intensive than simple drying. Cycle times can range from hours to several days depending on load, container, and formulation. Amorphous materials may require excipients that help preserve structure during freezing and drying. The method is widely used for biological materials, pharmaceuticals, and foods where heat drying would cause unacceptable change. Open questions remain about scaling cycles between laboratory and production equipment, and this gap affects technology transfer.
| Property | Value | Notes |
|---|---|---|
| Appearance | Porous, uniform cake or powder | Collapsed or shrunken cakes indicate process issues. |
| Reconstitution time | Seconds to several minutes | Depends on cake porosity, excipients, and diluent. |
| Residual moisture | 0.5-3% w/w | Product-specific; measured by Karl Fischer titration. |
| Typical storage temperature | 2-25 °C | Some biologics require 2-8 °C. |
| Container closure | Glass vial with elastomeric stopper | Sealed under vacuum or inert gas. |
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.
Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
The physics of freeze-drying couples heat transfer, mass transfer, and phase change. Heat supplied through the shelf must reach the sublimation front without melting the ice or degrading the product. Water vapor then travels through the already dried layer and leaves the chamber, where low pressure and cold traps keep it from returning. The dried layer acts as a resistance to vapor flow, so drying rate changes as the front recedes. Open questions remain about how pore structure and formulation heterogeneity affect drying uniformity at larger scales.
Lyophilization removes water from a frozen material by sublimation under reduced pressure. The process begins with freezing, which converts liquid water into ice and concentrates dissolved solids. Primary drying then lowers chamber pressure so ice changes directly into vapor without passing through a liquid phase. Secondary drying raises the shelf temperature to remove bound water that remains after ice sublimation. The result is a dry, porous structure that can be reconstituted later.
Formulation composition influences whether freeze-drying produces an intact cake or a collapsed mass. Excipients such as sugars and polymers can raise the collapse temperature and provide bulk during drying. The critical temperature for primary drying is often the collapse temperature or the glass transition temperature of the maximally concentrated phase. If the product temperature exceeds this threshold, the frozen matrix may soften and lose structure. Established practice therefore links shelf temperature and chamber pressure to the formulation's thermal properties.
Akt regulates TFEB, a master controller of lysosomal biogenesis, by direct phosphorylation of TFEB at serine 467. Phosphorylated TFEB is excluded from the nucleus and less active. Pharmacological inhibition of Akt promotes nuclear translocation of TFEB, lysosomal biogenesis and autophagy. Akt promotes G1-S phase cell cycle progression by phosphorylating and inactivating glycogen synthase kinase 3 (GSK-3) at Ser9. This prevents the phosphorylation and degradation of cyclin D1.
Epigenetics refers to the study of heritable changes in genetic activities that are distinct from changes in DNA sequences. Epigenetic analysis involves studying DNA-protein interactions, histone modifications, and DNA methylation. Linked-read sequencing has been used for studying DNA methylation patterns by many studies. For example, in 2021, a study investigated the DNA methylation differences in peripheral blood cells between twins, in which one twin had Alzheimer's Disease and the other was cognitively normal. Linked-read sequencing technology allowed researchers to identify more than 3000 differentially methylated regions between these twins discordant for Alzheimer's Disease, and investigation of these differentially methylated regions eventually led to identification of genes enriched in neurodevelopmental processes, neuronal signalling, and immune system functions
Take breast cancer as an example. The sensitivity of traditional ultrasound screening for breast cancer can be 76%. While with the blood-based mRNA diagnostic method, the sensitivity could be 80.6%. mRNA-based disease diagnostic technologies allow quantitive measurement of mRNA in the certain samples, such as leukemia As some technologies such as RNA-seq can provide the entire transcriptome of individual, the mRNA-based disease diagnosis can be developed in the landscape of personalized medicine. In HER-2 breast cancer, detection of ERBB2 mRNA expression levels is helpful in predicting response to anti-HER2-based treatments. As mentioned above, the mRNA-based disease diagnostic technology is more sensitive and specific to certain diseases. Even though there is no obvious symptom, the mRNA-based disease diagnostic technology can serve as screening method for early changes in RNA levels. High serum metadherin mRNA expression was observed in colorectal cancer and associated with poorly differentiated histological grades

90. ArXiv [Preprint]. 2026 Jul 29:arXiv:2605.17186v2. Operator splitting for exploiting linear-rate closure in solving infinite ODE hierarchies. Chang JC. We introduce an operator-splitting method for infinite hierarchies of linear ordinary differential equations (ODEs) indexed by nonnegative integers. When the coupling coefficients depend linearly on the count index, an exact transformation closes the equations on finite count-index windows without an upper-boundary value. For more general hierarchies, Strang splitting applies the linear-rate closure during the linear-rate substeps and a conventional capped solver to the remainder. We derive the closure from generating functions and the method of characteristics and extend it to multi-indexed systems. The derivation requires neither positivity nor mass conservation, so it applies to a wider class of systems than the examplar stochastic models presented here. We discuss branching processes, stochastic predator-prey dynamics, the Schlögl chemical kinetics model, and a telegraph model for gene expression. Through numerical experiments and computational cost analyses we demonstrate that our operator splitting method is typically advantageous for solving large scale systems in terms of memory usage and computational time, while retaining accuracy competitive with finite state projection (FSP) methods. Weather observation quality control systems verify probability, history, and trends. One of the main and simplest forms of quality control is the check of probability. This check throws out impossible observations, such as the dew point being higher than the temperature or data outside acceptable ranges, such as temperatures over 200 degrees Fahrenheit. Another basic quality control check is to have the data compared to preset geographic extremes, perhaps combined with diurnal variations. However this only flags the data as uncertain because the station could be reporting correctly but there is no way to know. A better way is to correlate with previous observations as well as the other simple checks. This method uses one hour persistence to check the quality of the current observation. This method makes continuity of observations better since the system is able to make better judgments on whether the current observations are bad or not.
Chromatin remodelers in the INO80 subfamily are made of multiple subunit complexes with split ATPase domains. The INO80 subfamily's protein domains are an N-terminus, two RuvB-like proteins (Rvb1 and Rvb2), and a C-terminus. The ATPase domain of the N-terminus functions in the identification of DNA damage and aids in the stability of telomeres. A long insertion in the ATPase domain is what recruits the Rvb1 and Rvb2 helicases. These helicases contribute to genome maintenance and are unique to the INO80 subfamily of chromatin remodeling complexes. This subfamily also contains an Arp4-actin complex that aids in stability of genes. The Arp5 subunit is required for ATPase function, binding to the DNA, and relocation of nucleosomes.
Although PI3K is the major mode of Akt activation, other tyrosine or serine/threonine kinases have been shown to activate Akt directly, in response to growth factors, inflammation or DNA damage. These can function even when PI3K activity is inhibited. Other studies have shown Akt can be activated in response to heat shock or increases in cellular Ca2+ concentration, via Ca2+/Calmodulin-dependent protein kinase kinase (CAMKK).

Corin converts the atrial natriuretic peptide (ANP) precursor, pro-ANP, to mature ANP, a cardiac hormone that regulates salt-water balance and blood pressure. In mice, corin deficiency prevents pro-ANP processing and causes salt-sensitive hypertension. Corin may also function as a pro-brain-type natriuretic peptide convertase. Corin-mediated ANP production in the pregnant uterus promotes spiral artery remodeling and trophoblast invasion. CORIN mutations have been reported in patients with preeclampsia. In mice, corin functions in the dermal papilla to regulate coat color in an Agouti-dependent pathway.
SNX8 contains a PX domain in its N-terminus, which is located between amino acids 71 and 181. A homology domain with yeast's PX domain is localized between amino acids 75 and 178 within this same domain. As it is a phosphoinositide-binding domain, it is important to highlight amino acids 109, 135 and 148 as residues directly related to phosphatidylinositol 3-phosphate since being specific binding sites, constituting a phosphoinositid binding site with a span of 40 amino acids. Furthermore, it includes a pair of phosphorylable tyrosines in positions 95 and 126 that are key in its function in the IFNγ-triggered IKKβ-mediated noncanonical signaling pathway. Overall, the PX domain main function is to target SNX8 mainly to early endosomes and other membranes rich in phosphatidylinositol 3-phosphate phospholipids.
A 1998 study was performed on NCAA football players during an 8-week spring training program. Those who took the ZMA capsules experienced greater increases in muscle strength. However, the study was funded by SNAC Systems Inc, which controls the trademark for ZMA. One of the study's authors, Victor Conte, has ownership equity in SNAC System Inc. In 2004, supplement company Cytodyne funded a study on 42 resistance-trained males using a product called Z-Mass PM that contained ZMA and additional ingredients Mucuna pruriens and polypodium vulgare. The study found that Z-Mass PM supplementation had no significant effects on total and free testosterone, IGF-1, growth hormone, cortisol, the ratio of cortisol to testosterone, or muscle and liver enzymes in response to training. No significant effects were observed in changes in strength, upper or lower body muscle endurance, or anaerobic sprint capacity. In another study done in 2006, a team of German scientists measured the effect of ZMA on testosterone levels. The study found that ZMA altered urine color due to increased secretion of zinc. It found no effect on testosterone levels.
Clostridium tetani produces tetanus toxin (TeNT protein), which leads to a fatal condition known as tetanus in many vertebrates (including humans). Tetanus toxin is produced from Clostridium tetani, a spore forming bacteria commonly found in soil, Tetanus is a paralytic disease that commonly affects newborns and non-immunized individuals. Tetanus enters the body of organisms through wounds or skin breaks and can be found in manure, soil, and dust. Tetanus mechanism includes tetanus preventing the transmission of glycine and γ-aminobutyric acid from inhibitory interneurons in the spinal cord, leading to activation of motor neurons causing contraction (known as spastic paralysis). It can also cause dysautonomia through a similar process that leads to autonomic dysinhibition. When tetanus toxin enters the body it is taken up by cholinergic nerve endings travel through axons into the spinal cord, the toxin is subsequently released from motor neurons and reuptaken into inhibitory nerve terminals triggering loss of inhibition and exaggerated reflexes in intoxicated individuals. Tetanus intoxication is a vaccine preventable illness.
The United States Food and Drug Administration and the European Medicines Agency have approved weight loss medications for adults with either a body-mass index (BMI) of at least 30, or a body-mass index of at least 27 with at least one weight-related comorbidity. This patient population is considered to have sufficiently high baseline health risks to justify the use of anti-obesity medication. The American Academy of Pediatrics had not previously supported the use of weight loss medication in adolescents but issued new guidelines in 2023, which recommend considering the use of weight loss medication in some overweight children aged 12 or older. The European Medicines Agency has approved semaglutide for children aged 12 or older who have a BMI in the 95 percentile for their age and a weight of at least 60 kilograms (130 lb). However, GLP-1 agonists may not be cost effective in this population.

No. Freeze-drying removes water but does not reliably kill microorganisms. Sterile lyophilized products are typically prepared aseptically before freezing or are sterilized by a validated method. Microbial control depends on the entire manufacturing process.
Storage temperature is set by the least stable component in the formulation. Proteins, vaccines, and some small molecules can degrade faster at higher temperatures. Refrigeration slows these changes but does not stop them completely.
Collapse occurs when the product exceeds its collapse or glass transition temperature during drying. The ice structure then loses support, and the cake may shrink, melt back, or become dense. Formulation and cycle adjustments are used to keep the product below that threshold.
Primary drying removes ice by sublimation under vacuum. Secondary drying removes water that is bound to the material, often by warming the product after most ice has left. Both stages occur below temperatures that would cause unwanted melting.
